Hand-made probes with Bacterial Artificial Chromosome
A Bacterial Artificial Chromosome (BAC) is a DNA construct based on a functional fertility plasmid (F-plasmid), used for transforming and cloning in bacteria, usually E. Coli. The F-plasmid contains the DNA fragment that has to be cloned (the usual insert size is 150-300kbp) and an antibiotic resistance gene. First, the plasmid is transfected into E. Coli cells, which are cultured with LB medium (Luria-Bertoni). Only the cells that incorporate the plasmid and, therefore, contain the antibiotic resistance gene will be able to grow. Finally, the cells are lysed and the cloned DNA sequence is isolated.
Human BAC libraries have played a critical role for the construction of physical maps and have also been used as intermediate substrates for genome sequencing. The mapped and sequenced BACs are useful resources for functional analysis of genes and for diagnostic assessment of chromosomal rearrangements by fluorescent in situ hybridization (FISH).
Human genome high-resolution BAC re-arrayed clone set (the “32k set”): this BAC clone set has been generated in collaboration between CHORI (Children's Hospital Oakland Research Institute) scientists and scientists at Marco Marra’s Genome Sciences Centre, Vancouver, BC Canada. This library contains 32.000 BAC clones that represent the mapped human genome.
BAC isolation as well as its labelling to make a hand-made probe can be requested as a service or can be delineated as a collaboration between research groups.
